Introduction of Exosomes Manufacturing in Cosmetics

Different types of extracellular vesicles (EVs) released from living cells present potential therapeutic benefits, such as microvesicles, apoptotic blebs, and exosomes. Among them, exosomes are released from multivesicular bodies (MVBs) rather than exocytosis. As a novel therapeutic platform, a suitable manufacturing process is required to produce exosomes cost-effectively at the appropriate scale. Here, we introduce our exosomes manufacturing strategies and their application in the transition of exosomes from laboratory scale to clinical production.

  • Upstream Processing of Cell Lines

Multiple cell lines retrieved from the patient or a working cell bank (WCB) can be expanded via the T-flask platform. To better achieve the potential that stem cell-derived exosomes can be used as drug candidates, upstream scalable manufacturing processes are necessary to produce large quantities of secreted exosomes. Additionally, serum-free media can be used to reduce contamination and aid in exosome purification.

  • Downstream Processing for Isolation and Purification

There are also significant downstream processing challenges for efficient exosomes isolation and purification. In general, four isolation methods are used which include size exclusion based on exosome diameter, sedimentation force or flotation density, precipitation-based nonspecific methods; and affinity-based capture. Ultracentrifugation is also the commonly used method to get a final pellet of the exosomes. Next, the most suitable approach would be used to obtain scalable and pure exosome products.

  • Exosome Analysis

For validation of exosome quality, we offer comprehensive characterization and quantification services. These methods include but are not limited to Nanoparticle Tracking Analysis (NTA), exosomal surface marker analysis (using FACS, WB, ELISA), physical observations (Mass spectrometry), and Tunable Resistive Pulse Sensing (TRPS). What’s more, in order to facilitate the storage and transportation of exosome products, we can also freeze-dry the isolated and purified exosomes.
