Released by almost all cell types, exosomes are small vesicular structures with sizes ranging from 40-150 nm in diameter. Nanosight nanoparticle tracking analysis is the technology to count and size nanoparticles in a liquid suspension. While the detection of the specific marker is not required and exosomes can be suspended in various solutions, it has been served as the best method for exosomes quantification. However, it is important to note that samples must b diluted to a specific range of concentrations to obtain accurate and reproducible size distribution and data.

Vesicle Flow Cytometry

Flow cytometry (FC) allows the detection of particles suspended in a fluid via the interaction with a laser beam. Flow cytometers often need to be customized to meet the needs of exosome detection. However, flow cytometry is not sensitive in the smallest size range and is also challenging to determine particle size

Surface Plasmon Resonance

Surface plasmon resonance (SPR) can detect the binding of exosomes to the surface through changes in the optical properties of the surface. The advantage of this technique is the ability to quantify subpopulations of exosomes or individual proteins present in exosomes.

Electron Microscopy

Electron microscopy (EM) is a gold standard to verify the quality of exosome preparations. However, the labor-intensive, low-throughput nature of this technique can easily lead to exosome loss resulting in a significant underestimation of exosome numbers.
