Introduction of Exosomes Purification in Cosmetics

Derived from extracellular vesicles (EVs), exosomes exist in a series of body fluids and play important roles in multiple physiological and pathological events. In recent years, there are different exosomes purification methods have been developed, such as ultracentrifugation and microfiltration, size exclusion chromatography (SEC), as well as immunoaffinity capture. To avoid loss of exosome fraction or damage to exosome membrane integrity during purification, optimization and improvement of current methods are required.   • Ultracentrifugation and microfiltration

While dilution of precious biological fluid samples can adversely affect many assays, it has a positive role as a concentration step during exosome purification. Compared with the isolation of exosomes from cell culture conditioned media, purification of exosomes from certain body fluids faces greater challenges due to the various impurities that may be present. A general strategy is to greatly dilute the biological fluid and increase the centrifugation duration prior to ultracentrifugation. Microfiltration utilizes a hydrophilized polyvinylidene fluoride membrane to purify exosomes from the samples.

  • Size Exclusion Chromatography (SEC)

Size-exclusion chromatography (SEC), also known as molecular sieve chromatography, is a chromatographic method to separate molecules in solution by their size. For exosome purification, vesicles can be separated from the smaller protein and nucleic acid contaminants.

  • Immunoaffinity Capture

A large number of proteins and receptors present in exosomal membranes can serve as the basis for immunoaffinity capture. Incubation of pre-concentrated extracellular vesicle samples with magnetic beads coated with antibodies against the target marker allows the specific isolation and purification of extracellular vesicles expressing the target on their surface.

Based on Tim4-immobilized magnetic beads, our developed exosome purification kit allows intact exosomes purification with higher purity compared with conventional methods. In addition, this method improves recovery efficiency, shortens reaction time, and reduces cytotoxicity.
