The Necessity of Microbial Identification

Microbial identification is a key aspect of cosmetic product safety and quality. Through the most basic morphological observation under the microscope, preliminary clues about whether they are gram-negative or gram-positive can be obtained. Then using commercial identification kits, such as API, the isolate can be identified.

In addition, the comparative gene sequence analysis of ribosomal DNA (rDNA), especially the 16S ribosomal RNA gene of bacteria or the LSU D2 region of fungi, has been proven to have the highest accuracy of microbial identification system technology.

Standard Methods for Microbial Identification

  • ISO 18416: Candida albicans
  • ISO 18416 provides the standard guidelines for the detection and identification of Candida albicans in cosmetic products. In general, the detection of Candida albicans is performed in a non-selective liquid medium (enriched broth), followed by the Candida albicans isolation on a selective agar medium. Due to the wide variety of cosmetics, this method may not have wide applicability. Depending on the level of detection required, other methods may be appropriate.

  • ISO 22718: Staphylococcus aureus
  • ISO 22718 provides the standard guidelines for the detection and identification of Staphylococcus aureus in cosmetic products. In general, the detection of Staphylococcus aureus is performed in a non-selective liquid medium (enriched broth), followed by the Staphylococcus aureus isolation on a selective agar medium. Due to the wide variety of cosmetics, this method may not have wide applicability. Depending on the level of detection required, other methods may be appropriate.

  • ISO 21150: Escherichia coli
  • ISO 21150 provides the standard guidelines for the detection and identification of Escherichia coli in cosmetic products. In general, the detection of Escherichia coli is performed in a non-selective liquid medium (enriched broth), followed by the Escherichia coli isolation on a selective agar medium.
