Introduction of Eye Irritation/Corrosion Test

During the development of cosmetic products, eye irritation evaluation is essential to avoid potential harm to the human body. As animal testing of cosmetics products is discouraged in the United States, while it is banned in Europe. The in vitro eye irritation test is a standardized acute eye irritation test method that can be used to determine the potential irritation of cosmetics.

  • Bovine Corneal Opacity and Permeability Test

The BCOP (Bovine Corneal Opacity and Permeability) assay is an in vitro eye irritation test method using living bovine corneal tissue. This assay directly assesses the effects of cosmetic products on the opacity and permeability of bovine cornea so that measures potential eye irritation. Theoretically, a range of parameters that relate to eye irritation can be obtained using this assay. For example, corneal opacity can be measured with an opacity meter and corneal permeability can be determined using sodium fluorescein.

  • Hens Egg Chorionallantoic Membrane Test

Hens egg chorioallantoic membrane (HET-CAM) test is an alternative test to the Draize rabbit eye test. In the test, the hatched eggs were carefully opened on the 9th day and the chorioallantoic membrane (CAM) was exposed. By visually inspecting the membrane through a microscope, an irritation score (IS) can be obtained and the test sample can be classified into no irritation, mild irritation, moderate irritation, and strong irritation.

  • Reconstructed Human Corneal Epithelial Models

Made from human-derived cells, reconstructed human corneal epithelial (RhCE) models have been applied for the eye irritation test (EIT). These cells are cultured on specially designed cell culture inserts to form a multilayer structure to mimic the corneal epithelium. Our in vitro RhCE models are suitable for eye irritation/corrosion tests of cosmetic products.

  • Red Blood Cell Test (RBC)

The Red Blood Cell (RBC) test is a novel method as an alternative to the Draize rabbit eye test. It is a cell-based cytotoxicity assay to examine ocular irritants by measuring lysis of erythrocytes (hemolysis) and denaturation of released oxyhemoglobin. Both hemoglobin leakage and oxyhemoglobin denaturation are measured and analyzed by spectrophotometry.
