The Necessary of Skin Irritation/Corrosion Test

In recent years, the cosmetics industry is developing rapidly, but it is also accompanied by stricter regulatory mechanisms. Of course, cosmetic manufacturers hope that their products will not irritate the skin. Unfortunately, many ingredients (such as surfactants, preservatives, emulsifiers, and many other organic substances) are inherently irritating.

As part of a basic cosmetic toxicology test program, the evaluation of the potential of a chemical that leads to skin irritation/corrosion must be assessed. Skin irritation refers to the reversible damage to the skin following the application of a test substance for up to 4 hours, while skin corrosion is defined as the irreversible damage to the skin following the application of a test substance for 3 mins up to 4 hours. Historically, rabbit is a powerful tool for cosmetics skin irritation/corrosion tests. However, due to differences in skin structure between humans and rabbits, as well as the cessation of animal testing of cosmetic ingredients specified in the Cosmetics Directive, alternative methods are urgently needed.

Introduction of RHE Models for Irritation/Corrosion Test

Nowadays, a series of alternative assays have been developed to determine the skin irritation/corrosion potential of cosmetics, especially the use of reconstructed human epidermis (RHE) in vitro skin models. In RHE skin models, keratinocytes play important roles in the initiation, modulation, and regulation of skin irritation. In this case, the functional stratum corneum allows the test of neat chemicals added directly to the stratum corneum. Compared with normal human skin, RHE models present higher penetration and sensitivity.
