The Introduction of Reproductive Toxicity

Reproductive toxicity is the process by which certain reproductive toxic substances interfere with normal reproduction in some way. It may adversely affect the sexual function and fertility of adult men and women, and cause developmental toxicity in offspring. According to reports, 3% of human newborns are affected by congenital anomalies, 5-10% of which are due to severe side effects of chemical compounds. Methods for Reproductive Toxicity Testing

The complexity of the reproductive system and the large number of tissue targets that induce deformities or postpartum effects provide a basis for the toxicity testing of chemicals in highly standardized and internationally recognized tests. Following OECD guidelines, there are both in vivo and in vitro reproductive toxicity tests. The in vivo tests are always more time-consuming, expensive, and raise ethical issues. In this case, various in vitro methods have been developed, such as whole embryo culture (WEC), Xenopus (FETAX) test, or chicken (CHEST).

Among them, the animal-free, cell-line-based embryonic stem cell test (EST) has been the most extensively studied test. Established from the inner cell mass, embryonic stem (ES) cell lines can be expanded under the control conditions and remain pluripotent. In the field of toxicology, the embryonic mouse stem cell has been used for reproductive toxicity tests.
