The Introduction of Phototoxicity Test

Phototoxicity, also known as photoirradiation, is defined as the toxic response induced upon initial exposure of skin to a test sample, and subsequent exposure to light. It is a type of photosensitivity requiring light but does not involve the immune system. In addition, it also involves toxic reactions induced by skin irradiation after systemic administration (oral, intravenous) of a chemical substance. If the chemical absorbs ultraviolet or visible light, the absorbed energy may cause molecular changes, causing the chemical to become toxic. For example, various synthetic compounds, including tetracyclines or fluoroquinolones, are known to cause these effects.

In Vitro 3T3 BRU Phototoxicity Test (OECD 432)

If a cosmetic product absorbs UV or visible light, it would be necessary to determine if it is likely to cause adverse phototoxic effects when intended for human use.

The in vitro 3T3 BRU Phototoxicity test is a sensitive approach to evaluate the potential of a chemical agent to induce phototoxicity. This test system contains the immortalized mouse fibroblast cell line, Balb/c 3T3. Using 3T3 cells, this method evaluates cytotoxicity by the uptake of neutral red after exposure to the test substance. This difference in absorption can be measured with a spectrophotometer, which also allows the distinction and quantification between viable, damaged, and dead cells. What’s more, this method has been adopted as OECD test guideline No 432 to predict acute phototoxicity effects in animals and humans in vivo.
